Therefore for creating relationships for existing records or updating a parent and maybe a single somehow special assoc it’s usually simpler to opt for other solution like manually updating those separately. The same is true for embeds, but as partial updates are not really a things on embeds at the db level as well it’s usually fine. It’s therefore only really useful if the whole set of assocs is handled (handling just subsets is possible but uncommon). This does not create “just an relationship” to an already existing record in the db. The existing data is then morphed to match the input by creating/updating/deleteing elements. Creating relationships #īoth cast_assoc/3 and put_assoc/4 compare preloaded assocs with the input provided. Schemas are used to map any data source into an Elixir struct. For changing values on an associations cast_assoc/3 or put_assoc/4, while for embeds it’s cast_embed/3 or put_embed/4. If this was all we had, we might have a few headaches come our way. Let’s take a look at a changeset that only casts: def changeset (user, attrs) do user > cast (attrs, :displayname,:email,:password) end. For changing fields on a schema you’d use cast/4, change/2 or put_change/3. Ecto.Changesets allow us to filter, cast, validate, and constrain the structs created by our schema that represents the database records. Therefore which set of function to go for is determined by what data you want to apply as change. ![]() For those functions values for fields are expected to already be “runtime format” values, Those can be applied by another set of functions on Ecto.Changeset: change/2, put_change/3, put_assoc/4 and put_embed/4. in an event sourcing system one might handle events, whose data has been validated before and is no longer expressed by constrainted data types. Ecto.Changeset exposes 3 functions for doing that when applying changes: cast/4, cast_assoc/3 and cast_embed/3.Ĭhanges are not always “user input”, which needs casting. The third type is the data type the runtime data is serialized into for database storage.Ĭonverting user input into the runtime data type is called “casting”. Then there’s the actual values one want’s to have have at runtime in schemas – your field :some_name, :date being a %Date. JSON serialize data might know a few more data types, but is still quite limited. in a web context forms often submit data with text values only. There’s user input, which is often provided in a subset of data types of what a schema is meant to carry. ![]() There are three conceptional types of data ecto deals with. To bring some clarity into the matter one first needs to understand a thing about the architecture ecto and changesets operate under. ![]() Especially the options around assocications are often problematic. People starting out with ecto are often confused by all the methods of getting changes applied to a changeset and therefore the database. ![]() This Lightpaper may be revised or altered the most current version supersedes previous versions. The Shroomies Light paper is a dynamic document and will be subject to continuous updates in response to changing legal and technical environments. The value of Shroomies NFTs should be based ONLY on their digital art value. Any and all utility defined in this Lightpaper cannot be guaranteed to Shroomies NFT holders and can be changed at any moment. Smart contract platforms and cryptocurrencies have captured mass attention but still have not been able to achieve mass adoption due to scalability and user experience issues. With respect to Shroomies NFTs, they are solely digital art and nothing more. Even on Ethereum, which is the most widely used smart contracts platform, there have not been many examples of DApps which have seen mass adoption. There have been a few cases where one or the other particular application temporarily succeeded in achieving a significant user base, but it led to crippling of the entire network during the high network load times. Essentially this means that even the most advanced and widely used platforms are not ready for mass adoption yet. On the other hand, there are a few smart contract platforms which boast of higher transaction throughput, but they compromise on decentralization in order to improve transaction speeds. Also, many of the upcoming solutions propose developing their own blockchains, neglecting the billions of dollars of market cap that DApps and other projects have already created on platforms like Ethereum and others. More importantly, they neglect the massive developer community and developer ecosystem that currently exists on platforms like Ethereum. Matic Network strives to solve the scalability and usability issues, while not compromising on decentralization and leveraging the existing developer community and ecosystem. It is an off/side chain scaling solution for existing platforms to provide scalability and superior user experience to DApps/user functionalities. The Matic development team has chosen Ethereum as the first platform to showcase its scalability and already has a working implementation for Ethereum on Kovan Testnet. It is expected to allow near-instant transfers, exchange and conversion of digital assets (e.g. crypto tokens) and cryptocurrencies in the future. The Matic Network is an adapted implementation of the Plasma framework for Ethereum to start with, but the “vision” of the Matic development team is to provide off/side chain scaling solutions for blockchains in general. Matic foundation intends to provide Matic wallet, payment APIs & SDKs, products, identity solutions and other enabling solutions that will allow developers to design, implement and migrate DApps built on base platforms like Ethereum. One of the key pillars that form the basis of Matic Network’s ideology is the improvement of user experience, this area is poorly developed for Blockchain applications as of now. The Matic Development team has already built high quality user experience Mobile/Web browser libraries which will enable businesses to create real world end user applications on a large scale. The development roadmap of the Matic Network also includes supporting cross-chain transfers and third-party Decentralized exchanges, liquidity pools etc. Great news! English paleontologist Charles Davies Sherborn in 1928 listed an 1835 series of articles by Agassiz as the first scientific description of the shark. English paleontologist Edward Charlesworth in his 1837 paper used the name Carcharias megalodon, while citing Agassiz as the author, indicating that Agassiz described the species prior to 1843. Swiss naturalist Louis Agassiz gave this shark its initial scientific name, Carcharodon megalodon, in his 1843 work Recherches sur les poissons fossiles, based on tooth remains. He described his findings in the book The Head of a Shark Dissected, which also contained an illustration of a megalodon tooth. This interpretation was corrected in 1667 by Danish naturalist Nicolas Steno, who recognized them as shark teeth, and famously produced a depiction of a shark's head bearing such teeth. The depiction of a shark's head by Nicolas Steno in his work The Head of a Shark DissectedĪccording to Renaissance accounts, gigantic triangular fossil teeth often found embedded in rocky formations were once believed to be the petrified tongues, or glossopetrae, of dragons and snakes. The shark's extinction coincides with a gigantism trend in baleen whales. A reduction in the diversity of baleen whales and a shift in their distribution toward polar regions may have reduced megalodon's primary food source. As the shark preferred warmer waters, it is thought that oceanic cooling associated with the onset of the ice ages, coupled with the lowering of sea levels and resulting loss of suitable nursery areas, may have also contributed to its decline. The animal faced competition from whale-eating cetaceans, such as Livyatan and other macroraptorial sperm whales and possibly smaller ancestral killer whales. Unlike the great white, which attacks prey from the soft underside, megalodon probably used its strong jaws to break through the chest cavity and puncture the heart and lungs of its prey. Juveniles inhabited warm coastal waters and fed on fish and small whales. ![]() It probably targeted large prey, such as whales, seals and sea turtles. The fossil record indicates that it had a cosmopolitan distribution. Megalodon probably had a major impact on the structure of marine communities. Their teeth were thick and robust, built for grabbing prey and breaking bone, and their large jaws could exert a bite force of up to 108,500 to 182,200 newtons (24,390 to 40,960 lbf). Extrapolating from a vertebral column and reconstructing a 3D model with dimensions based on all extant lamnid sharks suggests that a 16-meter-long (52 ft) individual may have been much larger than previous estimates, reaching an excess of 61.5 metric tons (67.8 short tons) in body mass an individual of this size would have needed to consume 98,175 kcal per day. Extrapolation from a vertebral centra with dimensions based on the great white shark suggests that a megalodon about 16 meters (52 ft) long weighs up to 48 metric tons (53 short tons), 17 meters (56 ft) long weighs up to 59 metric tons (65 short tons), and 20.3 meters (67 ft) long (the maximum length) weighs up to 103 metric tons (114 short tons). The most recent estimate with the least error range suggests a maximum length estimate up to 20.3 meters (67 ft), although the modal lengths are estimated at 10.5 meters (34 ft). Scientists differ on whether it would have more closely resembled a stockier version of the great white shark, the whale shark ( Rhincodon typus), the basking shark ( Cetorhinus maximus) or the sand tiger shark ( Carcharias taurus). While regarded as one of the largest and most powerful predators to have ever lived, the megalodon is only known from fragmentary remains, and its appearance and maximum size are uncertain. However, it is now classified into the extinct family Otodontidae, which diverged from the great white shark during the Early Cretaceous. It was formerly thought to be a member of the family Lamnidae and a close relative of the great white shark ( Carcharodon carcharias). Megalodon ( / ˈ m ɛ ɡ əl ə d ɒ n/ MEG-əl-ə-don Otodus megalodon), meaning "big tooth", is an extinct species of mackerel shark that lived approximately 23 to 3.6 million years ago (Mya), from the Early Miocene to the Pliocene epochs. ![]() You will receive another email once you have collected your order as confirmation. ![]() ![]() Computationally scoring these transitions determines the precise location and intensity of the RBP interaction and importantly, removes background from contaminating non-crosslinked RNAs derived from fragments of abundant transcripts ( 11, 12).Ī typical challenge for CLIP methods is the low yield of recovered RNA footprints that hovers in the nanogram range and complicates cDNA library construction efforts. Crosslinked 4SU preferentially pairs with guanosine during reverse transcription, which results in a characteristic T-to-C transition (or G-to-A when using 6SG) in the sequenced cDNA. In the case of 4SU, the photoaddition of reactive amino acids leaves a lesion at position 4 of the base and changes its base-pairing properties ( 14). The exocyclic thione group increases the photoreactivity of the base, allowing lower energy UV light (312 ≤ λ ≤ 365 nm) to be used for crosslinking. In PAR-CLIP, cells are incubated prior to crosslinking with modified nucleosides, 4-thiouridine (4SU) or 6-thioguanisine (6SG) that get incorporated into nascent RNAs. One widely used CLIP variant is photoactivatable ribonucleoside enhanced CLIP (PAR-CLIP) ( 13). (iv) Sequenced reads are aligned to the genome and high-confidence RBP binding sites are computationally separated from noise derived from fragments of abundant cellular RNAs ( 11, 12). (iii) RNA footprints are converted into cDNA suitable for next generation sequencing. (ii) The crosslinked ribonucleoprotein (RNP) of interest is immunopurified and RNA footprints are isolated. SELEX ( 9, 10), RNA Bind’n'seq ( 5), RNAcompete ( 4).Īll CLIP protocols share the same basic workflow: (i) RNA and interacting proteins are irreversibly crosslinked in intact cells by UV light and the RNA is digested to short footprints by RNases. This in turn reveals critically important details of RBP function, such as the location of its binding site relative to other cis-acting elements or its recognized sequence motifs, which often resemble but may not be identical to high-affinity motifs determined in vitro by e.g. In CLIP approaches, protected footprints of RNA irreversibly crosslinked to interacting RBPs are sequenced to precisely identify RBP binding sites on a target RNA. Crosslinking and Immunoprecipitation (CLIP) ( 7) and its many variants ( 8) address this need by allowing to identify not only the RNA bound by the RBP in living cells, but also its binding sites. RBPs typically recognize their targets at short, degenerate sequence-motifs ( 4–6), complicating prediction of RBP–RNA interactions and calling for systems-wide experimental methods to dissect the complex posttranscriptional gene regulatory networks controlled by RBPs. The importance of posttranscriptional gene regulatory networks is highlighted by the fact that >1500 human genes encode for proteins known or predicted to bind RNA ( 2) and that multiple diseases are associated with RBPs ( 3). RNA binding proteins (RBPs) participate in all processes involving RNA, including RNA biogenesis, processing, modification, translation and finally turnover. The fate of all eukaryotic RNAs is controlled by their protein partners which accompany them from transcription to turnover ( 1). These improvements cut the experimentation by half to 2 days and increases sensitivity by 10–100-fold. It is based on direct ligation of a fluorescently labeled adapter to the 3′end of crosslinked RNA on immobilized ribonucleoproteins, followed by isolation of the adapter-ligated RNA and efficient conversion into cDNA without the previously needed size fractionation on denaturing polyacrylamide gels. Here we present a streamlined protocol for fluorescence-based PAR-CLIP (fPAR-CLIP) that eliminates the need to use radioactivity. ![]() Crosslinking of 4SU or 6SG to interacting amino acids changes their base-pairing properties and results in characteristic mutations in cDNA libraries prepared for high-throughput sequencing, which can be computationally exploited to remove abundant background from non-crosslinked sequences and help pinpoint RNA binding protein binding sites at nucleotide resolution on a transcriptome-wide scale. One widely used CLIP variant is photoactivatable ribonucleoside enhanced CLIP (PAR-CLIP) that involves in vivo labeling of nascent RNAs with the photoreactive nucleosides 4-thiouridine (4SU) or 6-thioguanosine (6SG), which can efficiently crosslink to interacting proteins using UVA and UVB light. Crosslinking and immunoprecipitation (CLIP) methods are powerful techniques to interrogate direct protein-RNA interactions and dissect posttranscriptional gene regulatory networks. ![]() If any of these upcoming challenges take your fancy, get ahead of the curve and start creating!
